RNA methyltransferase-like 1 (METTL1) is known to induce m7G modification into RNA in eukaryote. To explorer the developmental role of m7G, we generated Drosophila METTL1-KO strain and found the mutant shows male fertility defect. In METTL1-KO, mitotic divisions of germline cells are impaired and no elongated spermatid are observed. Biochemical analyses show METTL1 induces m7G into tRNAs in vitro and in vivo, and regulates expression level of m7G target tRNAs, suggesting that the regulation of tRNA stability by METTL1 links to Drosophila fertility via m7G modification.
