Formation of pericentromeric heterochromatin via ZNF518s that link satellite DNA to heterochromatin

Heterochromatin present at pericentromeric alpha-satellites is essential for proper chromosomal segregation. Here, we demonstrate a novel mechanism by which ZNF518 plays central roles in heterochromatin formation at pericentromeres. We investigate this mechanism by determining the cellular localization of ZNF518, centromeric proteins and heterochromatin factors. The results indicate that the multiple segments within ZNF518 contributes to their interactions with CENP-B, HP1, and G9a histone methyltransferase. This study identifies a novel mechanism underlying pericentromeric heterochromatin formation mediated by the central component ZNF518.
